In this advanced high school biotechnology unit, developed by ABE Greater Los Angeles Master Teacher Fellow Dan Baker, students explore commercial protein purification techniques by engineering a plasmid to include a hemagglutinin (HA) affinity tag for red fluorescent protein (RFP). Through a blend of bioinformatics, primer design, and molecular biology techniques, students simulate and perform the steps needed to create a tagged protein ready for purification using chromatography.
